PPARγ对鸡Perilipin1基因的转录调控研究

哺乳动物的研究结果表明,脂滴包被蛋白(Perilipin1)参与脂肪细胞的分化和糖脂代谢的调节,与肥胖的发生、发展密切相关。与此类似,鸡Perilipin1在脂肪细胞脂肪分解的过程中也发挥着重要的调控作用,但其确切的转录调控机制尚未阐明。小鼠的研究表明,Perilipin1基因是过氧化物酶体增殖物激活受体γ(PPARγ)的一个靶基因,其转录主要受PPARγ的调控。鉴于此,本文通过报告基因、凝胶阻滞等方法分析了PPARγ对鸡Perilipin1基因的转录调控作用。报告基因的结果表明,PPARγ对鸡Perilipin1启动子-1992/-11区域的转录活性没有显著影响;凝胶阻滞的结果表明,PPARγ/RXRα蛋白不能够与鸡Perilipin1启动子-1912/-1882区域结合。结论:鸡Perilipin1基因的-1992/-11区域不存在PPARγ的结合位点。

Studies in mammals have shown that, Perilipin1 was involved in adipocyte differentiation and regulation of glucose and lipid metabolism, and was closely related to the occurrence and development of obesity. Similar to this, chicken Perilipin1 plays important roles in regulating the process of adipocyte lipolysis, however,its transcriptional regulation mechanism in poultry has not been illuminated.Studies in mice have shown that Perilipin1 is a target gene of peroxisome proliferator activated receptor gamma (PPAR). The transcription of Perilipin1 gene is mainly regulated by PPARγ. In the present study, the report gene assay and electrophoretic mobility shift assaywere used to reveal the chicken Perilipin1 gene transcriptional regulation mechanism by PPARγ.Report gene results indicate that PPARγ could not activate chicken Perilipin1 gene promoter region from -1992 to -11. And the result of EMSA showed that PPARγ/RXRα could not bind to the -1912/-1882 region of chicken Perilipin1 gene promoter.Conclusion: there is no PPARγ binding site in the -1992/-11 region of chicken Perilipin1 gene.
