目的:探讨白藜芦醇对长波紫外线(Ultraviolet A, UVA)辐射后人体成纤维细胞增殖活性及炎性因子的影响。方法:取第四代对数生长期的人体成纤维细胞,用MTT法检测0.01mmol/L、0.1mmol/L、0.5mmol/L、1.0mmol/L白藜芦醇分别处理后6h、24h、48h、72h细胞增殖活性,筛选出适宜的浓度。以10J/cm2UVA辐射细胞后立即加入白藜芦醇,继续培养6h、24h、48h、72h,MTT法检测细胞的增殖活性,ELISA法检测上清液中IL-1α、IL-1β、IL-6蛋白水平。结果:0.01mmol/L、0.1mmol/L白藜芦醇对正常人体成纤维细胞增殖活性无明显影响。0.01mmol/L白藜芦醇对UVA辐射后细胞增殖活性有明显的保护作用,对IL-1α、IL-1β、IL-6的分泌有明显抑制作用。结论:0.01mmol/L白藜芦醇对UVA辐射后的成纤维细胞有光防护作用,这种作用是通过保护细胞增殖活性及抑制细胞因子IL-1α、IL-1β、IL-6分泌实现的。

Objective: To investigate the effects of resveratrol on ultraviolet A (UVA)-induced cellular proliferation and inflammatory cytokines of human fibroblast. Method: The fourth generation human fibroblasts in logarithmic growth phase were used. Fibroblasts, which were respectively co-cultured with resveratrols in the concentrations of 0.01mmol/L, 0.1mmol/L, 0.5mmol/L, 1.0mmol/L for 6h, 24h, 48h, 72h, were tested using MTT to screen the optimal concentration. Immediately after irradiation with 10J/cm2UVA, the fibroblasts were treated with resveratrols and cultured for 6h, 24h, 48h, 72h. Cellular proliferation was tested using MTT, and the levels of IL-1α, IL-1β, IL-6 were measured using ELISA. Results: 0.01mmol/L and 0.1mmol/L resveratrols showed no influence on the cellular proliferation of normal human fibroblasts. 0.01mmol/L resveratrol prsent obviously protective effects on UVA-induced inhibition of cellular proliferation and secretion of IL-1α, IL-1β, IL-6. Conclusion: Resveratrol in 0.01mmol/L has photo-protective effects on UVA-induced fibroblasts, and the procedure is closely related to cellular proliferation and cytokines IL-1α, IL-1β, IL-6 secretion.
