自交不亲和是导致柑桔无籽的重要原因之一。无籽沙糖桔是本课题组从沙糖桔自交亲和芽变中选育而成的自交不亲和品种,是研究柑桔自交不亲和性/自交亲和性的宝贵材料。然而,在这个过程中究竟有哪些基因参与了调控而导致了SI的发生,目前还不清楚。本研究利用表达谱芯片技术对8个SSH文库进行筛选,共获得了1830个上调或下调的差异克隆。与沙糖桔相比,胚胎发育(Embryonic development)、泛素途径(Ubiquitination pathway)、钙信号途径(Ca2+-signaling pathway)和赤霉素4种途径中的5个基因显著上调表达。Auxin transporter protein 1在无籽沙糖桔柱头中明显上调表达;Gibberellin receptor在无籽沙糖桔花药和子房中高表达,推测两个基因可能作为泛素/26S蛋白小体降解途径中激素应答因子参与了无籽沙糖桔SI反应。
Self-incompatibility (SI) is an important trait of Citrus plants that is exploited by farmers for producing seedless fruit in Citrus. However, the molecular mechanism of SI in Citrus is not well understood. 'Wuzishatangju' (Citrus reticulata Blanco) (Self-incompatibility, SI) is an excellent seedless mandarin cultivar derived from a bud sport variation of a seedy 'Shatangju' cultivar (Self-compatibility, SC). Therefore, the two cultivars are excellent materials to study the mechanisms of SI and/or SC in Citrus. An integrative strategy combining eight suppression subtractive hybridization (SSH) libraries with cDNA microarray was used to study the different molecular mechanisms between SI 'Wuzishatangju' and SC 'Shatangju' (control) mandarins. A custom microarray screen resulted in a total of 1820 up- or down-regulated (Ratio≥3.0) obtained from 9810 positive clones. The expression of genes involved in embryonic development, ubiquitination pathway, Ca2+-signaling pathway, gibberellins (GAs) and auxin was significantly up-regulated in SI 'Wuzishatangju' compared with SC 'Shatangju' mandarin. The microarray analysis suggests that ubiquitin mediated proteolysis pathway might be involved in the SI reaction of 'Wuzishatangju.' Our research highlighted some candidate pathways that participate in the mandarin SI/SC reaction and could be beneficial for understanding the SI/SC systems evolution and breeding seedless citrus fruits in the future.
