目的 针对小鼠凋亡相关蛋白3制备多克隆抗体。方法 合成针对小鼠APR3 cDNA文库对应翻译的氨基酸序列并加保护后藕联载体免疫2只新西兰纯种大白兔, 获取免疫血清共50ml, 经蛋白 A 柱 纯化及 抗原抗体亲和 纯化后, 采用Western-blot及免疫细胞化学染色进行特异性鉴定。结果 构建了针对小鼠APR3 cDNA文库所对应两种序列所对应的抗体,经检测证实其中一个抗体Western blot证明能特异性性识别小鼠组织及NIH-3T3细胞。结论 成功制备了能特异性识别小鼠APR3的多克隆抗体。

PR3 is a novel protein with unknown function. Good antibody is essential to its function study. Currently there is no commercial antibody against mouse APR3. To develop antibody against mouse APR3, we synthesized two peptides of APR3 to immune rabbit. The serums from immunized rabbits were further purified by affinity chromatography. The specificity was checked by Western Blot and immunity, which demonstrated a band at the predicted molecular weight. In conclusion, an antibody specific to mouse APR3 was developed.
