电针对糖尿病小鼠骨骼肌纤维类型分化的影响

目的:探讨电针对糖尿病小鼠骨骼肌纤维类型分化的影响。方法:将45只C57小鼠采用高脂饮食联合小剂量STZ诱导2型糖尿病小鼠模型,随机分为对照组,药物组和电针组,每组15只。电针组小鼠取"后三里"、"内庭"给予电针治疗,药物组用二甲双胍(250mg/kg体重)灌胃,对照组用生理盐水(2ml/kg体重)灌胃,均1次/日,连续治疗28天。取比目鱼肌检测骨骼肌类型标志肌球蛋白重链(myosin heavy chain,MHC) mRNA和蛋白的表达情况。结果:与药物组及对照组相比,电针治疗后MHCI mRNA及蛋白的表达明显增高(P <0.01),而MHCIIb mRNA及蛋白的表达明显下降(P <0.01)。结论:电针可以诱导糖尿病骨骼肌MHCI的表达,促进糖尿病骨骼肌向I型纤维转化。

Objective: To observe the effect of electro-acupuncture on skeletal muscle fiber transformation in diabetic mice. Methods: 45 C57 mice were randomly divided into control group, drug group and EA group, with 15 mice in each group. The high fat diet combined with small dose of STZ was used to induce the model of type 2 diabetes. Mice in the EA group were treated by electro-acupuncture in point "Housanli" and "Neiting", drug group mice were lavaged with metformin (250 mg / kg body weight) , mice in control group were lavaged with normal saline (2ml / kg body weight), once daily for 4 weeks. Soleus muscle was taken to detect the expression of mRNA and protein of MHC, a marker of skeletal muscle type. Results: Compared with the drug group and the control group, the expression of mRNA and protein of MHCI in mice with electro-acupuncture was significantly increased (P < 0.01), while the expression of mRNA and protein of MHCIIb was significantly decreased (P < 0.01). Conclusion: Electro-acupuncture can induce the expression of MHCI in diabetic skeletal muscle , and promote the transformation of skeletal muscle to type I fiber.
