目的:为山楂叶提取物建立专属性含量测定方法。方法:以芦丁为对照品,采用紫外-可见分光光度法,在500nm波长处测定总黄酮的含量;以金丝桃苷为对照品,采用高效液相色谱法,用C18色谱柱,以乙腈-甲醇-四氢呋喃-0.5%冰醋酸溶液(1:1:19.4:64.2)为流动相,检测波长为363nm,测定金丝桃苷的含量。结果:芦丁浓度在0.0085-0.0511mg/mL范围内,与吸光度线性关系良好,相关系数为0.9997,平均回收率为98.95%,RSD=1.02%(n=9);金丝桃苷含量在0.0888~0.4444μg范围内,与峰面积线性关系良好,相关系数为0.9995,平均回收率为99.24%,RSD=1.21%。结论:该法操作简便、准确,可作为山楂叶提取物的质量控制。

Object:To establish a special determination method for folium crataegi extract.Methods:Rutin was reference substance and the concentration of flavones was determined by UV-VIS spectrosc- opy at the wavelength of 500nm;Using HPLC to determine the content of hyperoside with C18 column, acetonitrile-methanol-tetrahydrofuran-0.5% glacial acetic acid solution (1:1:19.4:64.2) as the mobile phase,the detective wavelength was 363nm.ResuLts:The linear range of calibration curve was 0.0085-0.0511mg/mL for the rutin(r=0.9997),the average recovery was 98.95% and the RSD was 1.02%(n=9);The linear range of calibration curve was 0.0888~0.4444μg for the hyperoside (r=0.9995),the average recovery was 99.24% and the RSD was 0.24%(n=9).Conclus- ion:The method is simple and accurate with a good reproducibility and can be used for the quality control of folium crataegi extract.
