补肾活血中药对高糖条件下纯化培养的视网膜神经节细胞Glu释放量的影响
Effect of bushenhuoxue serum on the Glureleaseof purified retinal ganglion cells in high glucose level conditions
本文探讨了稳定高糖及糖波动条件对体外纯化培养的视网膜神经节细胞(retinal ganglion cells,RGCs) 谷氨酸(Glutamate, Glu)释放量的影响,以及补肾活血(bushenhuoxue)中药的干预作用。采用抗体联合两步纯化法培养新生(Sprague –Dawley, SD)大鼠RGCs,将其分别置于模拟正常条件、稳定高糖条件及糖波动条件下进行培养,并以补肾活血中药复方含药血清进行干预。实验分为正常对照组、正常中药干预组,稳定高糖组、稳定高糖中药干预组,糖波动组、糖波动中药干预组;分别在试验干预24h、48h及72h后,用全自动氨基酸分析仪测定各组细胞外液中Glu的含量, SPSS13.0对所得数据进行统计学分析。结果显示糖波动条件持续24 h ,RGCs的Glu 释放量较正常条件及稳定高糖条件下均明显增加(P<0.05);补肾活血中药血清能降低24 h、72 h段正常条件及糖波动条件下RGCs的Glu 释放量,降低各时段稳定高糖条件下的Glu 释放量(P<0.05)。 本实验表明糖波动条件能明显增加RGCs的Glu释放量,导致大量Glu在细胞外聚积,细胞外高浓度Glu聚积最终引发RGCs神经兴奋性毒性,加剧细胞损害;补肾活血中药能在一定程度降低稳定高糖及糖波动条件下RGCs的Glu释放量,减轻Glu神经兴奋性毒性作用对RGCs的损伤,这可能是补肾活血中药防治DRP的药物干预途径之一。
o investigate the effect of serum with TCM for bushenhuoxue on the Glu release of purified retinal ganglion cells in high glucose level conditions Methods: Two monoclonal antibodies,anti-rat SIRP(OX-41)against rat macrophage and antibody against rat Thy-1.1(OX-7) were used to purify and characterize RGCs from rat neonates by means of two-step filtration. Purified RGCs were cultured in steadily high glucose conditions﹑glucose fluctuated conditions and normal control conditions ,bushenhuoxue medicine-containing serum was added to the culture medium. The Experiment was assigned into normal control group﹑TCM interventing group﹑steadily high glucose group﹑steadily high glucose group intervened with TCM﹑glucose fluctuated group﹑glucose fluctuated group intervened with TCM; Deteced the content of the glutamate(Glu) in the extracellular fluid with a full- automatic biochemical analyzer. And then the data obtained was treated with SPSS 13.0. Results:The release of Glu was obviously increased in glucose fluctuated conditions at 24 hours(P<0.05), and the release of Glu was obviously decreased by TCM intervention both in steadily high glucose conditions at each period and glucose fluctuated conditions at 24 hours and 72 hours(P<0.05). Conclusion: Bushenhuoxue therpy could decreas the Glu release from RGCs cultured in both steadily high glucose conditions and glucose fluctuated conditions, accordingly it might be one of the important mechanisms of bushenhuoxue therpy could prevent and cure DRP.
