柑橘木虱Diaphorina citri Kuwayama可培养性内生细菌的分离鉴定

本文通过采用四因素三水平L9(34)的方法 对柑橘木虱可培养内生细菌分离培养基进行优化,结果表明,在各种试验操作条件、培养条件一致的情况下,在所设计的各种影响因素、水平中,培养基为TSB、pH 值7.0、血淋巴为柞蚕血淋巴、生理盐水浓度为0的培养基可以分离出最大可能多的柑橘木虱内生细菌。将分离到的柑橘木虱内生细菌通过16S rDNA的方法进行鉴定,结果标明柑橘木虱内生可培养细菌分别为木糖葡萄球菌(Staphylococcus xylosus)、葡萄球菌属(Staphylococcus sp.)、芽孢杆菌属(Bacillus sp.)、肠杆菌属(Enterobacter sp.)、短小杆菌属的Curtobacterium oceanosedimentum、砖红色微杆菌(Microbacterium testacyeum)、嗜麦芽寡养单胞菌(Stenotrophomonas maltophilia)、柠檬色短小杆菌(Curtobacterium citreum)。生理生化鉴定结果表明内生细菌部分常见的生理生化特征与16S rDNA序列鉴定结果基本一致。
In this paper, by using four factors and three levels L9 (34) method , the medium can be optimized to separation the endophytic bacteria of Citrus psyllid ,The results show that a variety of test operating conditions, consistent with the culture conditions, various factors influence the design level, the medium of TSB, pH 7.0, for the hemolymph hemolymph pernyi, saline concentration of 0 the medium can be isolated within the largest possible number of bacteria in raw citrus psyllid.The bacteria subsequently identified based on their 16S rRNA genes.They were Staphylococcus xylosus, a possible novel species in Staphylococcus sp., Bacillus sp., Enterobacter sp., Curtobacterium oceanosedimentum, Microbacterium testacyeum, Stenotrophomonas maltophilia, Curtobacterium citreum. The results showed that the physiological and biochemical identification of endophytic bacteria some common physiological and biochemical characteristics consistent with the 16S rDNA sequence identification.
