目的 探讨三氧化二砷(Arsenic trioxide,As2O3)对动脉平滑肌细胞(Smooth muscle cell,SMC)的细胞生物学作用。方法 进行家兔主动脉SMC的原代培养,确定As2O3对动脉SMC抑制的浓度界限,应用台盼蓝染色法研究As2O3对动脉SMC活力的影响。同时应用透射电镜观察As2O3引发的SMC病变,以及应用3'末端DNA片段原位标记法(Terminal uridine nick-end labeling,TUNEL)研究As2O3诱导SMC的凋亡。 结果 在本研究中,As2O3对家兔主动脉SMC增殖具有抑制作用,并呈浓度和时间依赖性,当As2O3浓度为3.39和4.52μmol/L时对家兔主动脉SMC增殖抑制尤为显著。电镜和TUNEL结果显示,As2O3组的SMC出现了病理性细胞坏死和凋亡的特征性变化,这两种形式同时存在。与对照组比较,凋亡细胞显著增多(P<0.001)。结论 在一定浓度范围内,As2O3对SMC有明显的抑制作用,主要表现为细胞凋亡。

Objective To explore the biology effect of As2O3 on smooth muscle cell (SMC). Methods SMCs isolated from aorta of rabbit were cultured. SMC was inhibited by As2O3, the concentration scope of which was confirmed. The trypan viability test was used to test the viability of SMC affected by As2O3, the transmission electron microscope (TEM) was used to observe the change and TUNEL was used to detect the apoptosis induced by As2O3. Result The proliferation of SMC was depressed by As2O3, which may rely on the dose and time. The inhibiting effect showed significantly when As2O3 was at the concentration of 3.39 and 4.52μmol/L. The TEM and the TUNEL results confirmed that the SMC cultured with As2O3showed pathological necrosis or apoptosis. There was an important difference between the control group and the As2O3 group (P<0.001). Conclusion SMC was inhibited by As2O3 in a dose scope and the main form was apoptosis.
