对AGCU Database Y24试剂盒在3130遗传分析仪上采用POP-7?聚合胶进行电泳时出现在DYS391上的非特异峰进行分析

近几年，越来越多的商业化Y-STR试剂盒被广泛运用于法医遗传学的研究。本课题组在采用AGCU Database Y24对中国西南地区汉族人群进行群体调查时，发现当ABI PRISM 3130遗传分析仪使用POP-7?聚合胶对该试剂盒进行电泳分析时，在等位基因座DYS391的分型区内总会出现一个非特异峰，而这可能会对数据的分析带来一定的干扰。当我们采用POP-4?聚合胶重复该实验时，该非特异峰则出现在DYS391的分型区外，不会影响DYS391的数据分型。本文将对这一特殊现象进行报道和初步分析，以提醒学者们采用AGCU Database Y24进行调查研究时，应注意电泳分析仪上聚合胶的选择。?????
urrently, lots of commercially available Y-STRs kits have been applied widely in forensic genetics. Also, we applied the AGCU Database Y24 kit into the population data. Interestingly, an abnormal peak at DYS391 was observed when using POP-7? polymer on ABI PRISM 3130 Genetic Analyzer, which may be a potential risk of wrong genotyping. Then, we repeated the experiment using the POP-4? polymer. The abnormal peak was appeared far away from the typing zone. Here, we reported this phenomenon to remind the users of AGCU Database Y24 attention to the type of polymer.
