目的 探讨小泛素相关修饰蛋白2/3(Small ubiquitin-related modifier 2/3,SUMO2/3)诱导肝癌细胞凋亡的可能机制。方法 体外培养人肝细胞癌细胞株SMMC-7721,免疫荧光染色观察SUMO2/3蛋白的表达,免疫荧光双标观察过表达或敲低SUMO2/3与活化的caspase-3的关系;利用Western Blot法检测细胞凋亡标志性蛋白caspase-3和内质网(Endoplasmic reticulum, ER)应激相关蛋白的变化。结果 SUMO2/3蛋白在肝癌细胞的细胞核中稳定表达,ER 应激诱导剂衣霉素(Tunicamycin, TM)刺激后SUMO2/3表达量增加。过表达SUMO2/3可诱导肝癌细胞凋亡,通过检测ER 应激相关蛋白的水平,发现肝癌细胞过表达SUMO2/3上调了Bip、p-PERK、p-EIF2α、ATF6、ATF4、CHOP蛋白水平。结论 SUMO2/3可能是通过ER途径诱导肝癌细胞凋亡。?????

Our previous study showed that SUMO2/3 over-expression inhibited the proliferation of hepatoma cells, but the mechanism is still unknown. In this paper, we observed SUMO2/3 expression and found that endoplasmic reticulum (ER) stress inducer Tunicamycin (TM) increased SUMO2/3 protein levels in the nuclei of hepatoma cells. Meanwhile, we showed that SUMO2/3 over-expression increased cleaved caspase-3, Bip, p-PERK, p-EIF2α, ATF6, ATF4 and CHOP levels. These results suggested that SUMO2/3 induces apoptosis of hepatoma cells through ER pathway.
