目的 探讨CXCL12对人早孕期蜕膜基质细胞(DSCs)的生物学调节作用,解析CXCL12/CXCR4信号通路在母-胎对话中的作用机制。方法 收集人早孕期蜕膜及绒毛组织,分别分离培养DSCs及滋养细胞;收集滋养细胞条件培养液(TCM);侵袭实验分析CXCL12及TCM对DSCs体外迁移、侵袭能力的影响。结果 CXCL12和TCM可明显促进人早孕期DSCs体外迁移与侵袭,并且CXCL12或CXCR4的中和抗体可有效抑制CXCL12及TCM对DSCs的促侵袭作用。结论:滋养细胞通过CXCL12/CXCR4信号通路以旁分泌方式促进人早孕期DSCs迁移与侵袭,提示CXCL12/CXCR4在母-胎免疫调节中起重要作用。

Objective To explore the role of CXCL12/CXCR4 axis on the motility of human first-trimester decidual stromal cells (DSCs). Methods Human first-trimester decidual and villous tissues were collected for DSCs and trophoblasts isolation, respectively. Then the conditioned culture medium from trophoblasts (TCM) was prepared and stored. An in vitro invasion assay was conducted to estimate the change of DSCs invasiveness after treated with exogenous CXCL12 or TCM. Results Both CXCL12 and TCM significantly increased the number of migrated DSCs, and CXCL12 or CXCR4 neutralizing antibody effectively abolished the enhanced invasion of DSCs induced by CXCL12 or TCM. Conclusion Both CXCL12 and TCM are able to improve the invasive ability of human first-trimester DSCs via CXCR4 ligation, which contributes to the crosstalk between trophoblasts and DSCs.
