小鼠乳腺中胰岛素样生长因子及其受体的表达
Expression of Insulin-like Growth Factors and Insulin-like Growth Factor Receptor in Mouse Mammary Gland
选取小鼠乳腺发育过程中的若干时点,采用间接免疫荧光的双重染色方法,通过激光扫描共聚焦显微技术系统检测乳腺中胰岛素样生长因子(IGFs)及其受体(IGF-IR)的表达,并进行定位和定量研究,结果显示:⑴ IGF-I对青春期乳腺发育和妊娠期初始阶段的导管分支和小叶腺泡形成有重要作用,对泌乳期启始阶段几乎不起作用,但对维持泌乳期泌乳有一定影响;IGF-I在各个发育阶段主要弥散地分布在导管、腺泡的上皮细胞、脂肪细胞的核旁胞质区域和基质;⑵ IGF-II在乳腺导管发生和腺泡发育过程中发挥作用,尤其对妊娠期小叶腺泡形成和分化有显著作用;IGF-II在泌乳期乳腺大量表达也表明它能促进和维持泌乳。⑶ IGF-IR在整个乳腺发育过程中表达,尤其是妊娠期和泌乳后期高水平表达,主要存在于导管和腺泡上皮细胞、脂肪细胞和基质,介导IGFs促进导管和腺泡发育,维持泌乳的作用。
In this research, some time points were set in all stages of mouse mammary gland development Double staining mammary tissue slices was examined using a laser scanning confocal microscope. The results of localization and quantification of Insulin-like growth factors and their receptor(IGF-IR) revealed that: ⑴ IGF-I played a vital role on duct branching and lobuloalveolar morphogenesis during virgin and early pregnant mammary gland, and useless to initiate lactation but useful to maintain lactationg; ⑵ IGF-II played a key role on duct morphogenesis and alveolar development, especially was important to lobuloalveolar morphognesis and differentiation during pregnant mammary gland. The high level expression of IGF-II in lactating mammary gland appeared to be critical for promoting lactation and maintaining milk production.⑶ IGF-IR expressed in all stages of mammary gland development, especially highly expressed in pregnance and later lactaion, distributed in ducts, alveolar, matrix and adipocytes, which displayed that IGFs promoted ductal morphogenesis and alveolar development, maintained lactation by IGF-IR.
