N型乙酰胆碱受体α10亚基在大鼠内耳耳蜗、前庭中的表达
Expression of a10 Acetylcholine Receptor subunit mRNA in the Rat Cochlea and Vestibular Inner Ear
目的:研究N型乙酰胆碱受体α10亚基在大鼠前庭终器、Scarpa’s 神经节、基底膜、螺旋神经节中的表达。方法:运用RT-PCR技术分别检测大鼠前庭终器、Scarpa’s 神经节、基底膜、螺旋神经节编码N型乙酰胆碱受体α10亚基的基因表达。以大鼠脑组织RNA为阳性对照。结果:在前庭终器和基底膜上扩增出标志N型乙酰胆碱受体α10亚基基因表达的PCR产物。结论:N型乙酰胆碱受体α10亚基在大鼠前庭终器、基底膜上有表达,在Scarpa’s 神经节、螺旋神经节中无表达。为乙酰胆碱是内耳重要的神经递质之一提供了支持,并为更完整的理解N型乙酰胆碱受体在内耳中的传入-传出调节中的作用提供了依据。
objective: To investigate the expression of the α10 acetylcholine receptor subunit mRNA in the rat inner ear vestibular end-organ, Scarpa’s ganglion ,cochlea basement membrane and spiral ganglion. Method: Using a combination of reverse transcription follow by polymerase chain reaction, expression of the α10 acetylcholine receptor subunit mRNA in the rat inner ear vestibular end-organ, Scarpa’s ganglion ,cochlea basement membrane and spiral ganglion was examined. The brain RNA was used as positive control. Result: PCR amplification products representing subunit gene expression of the α10 acetylcholine receptor subunit was amplified in vestibular end-organ and cochlea basement membrane. Conclusion: The result showed thatα10 acetylcholine receptor subunit mRNA is expressed in the rat vestibular end-organ and cochlea basement membrane, but in the Scarpa’s ganglion and spiral ganglion, indicating that acetylcholine is one of the important neurotransmitters in the vestibular system, and provide a more complete understanding of the role of nAChR in the neurochemical transmission subserving the efferent-afferent interacation in the inner ear.
