融合疫苗在NOD/XCID人免疫重建鼠中抗肾癌的作用

目的:通过人的脐血造血干细胞移植于NOD/SCID小鼠中重建人的免疫系统,并研究DC融合疫苗在该重建免疫小鼠体内抗肿瘤的机制,探讨该DC融合疫苗抗肿瘤的效应和价值。 方法:分别将NOD/SCID小鼠分为ABCDEF6组,其中BCDEF5组重建人免疫系统,之后CDEF4组于皮下构建A498(肾癌细胞)荷瘤模型,之后CDE三组分别给与CD,GM-CSF修饰的融合细胞,GM-CSF-B7.1修饰的融合细胞用以治疗肿瘤模型,随后监测各组之间免疫功能的变化和成瘤模型的大小。 结果:免疫构建组的老鼠均监测到了人免疫细胞:CD3,CD45,CD56,且人免疫细胞数量随时间延长而升高,在第8周达到顶峰,之后逐渐下降,免疫重建组单纯纯移植肿瘤后人免疫细胞数量与单纯的免疫重建组相比是降低的,但之后应用了DC融合疫苗治疗,提高人免疫细胞数量,且DC融合疫苗明显降低了肾癌肿瘤组织的大小。 结论:确定了DC融合疫苗具有增强免疫能力,并且具有强烈的抗肿瘤作用,并证明了特定修饰对融合疫苗抗肿瘤效应有明显提高。
Objective: through human cord blood stem cell transplantation in NOD / SCID mice to reconstituted human immune system, and to study the mechanism of DC fusion vaccine anti-tumor in immunized reconstruction mice , and to explore the DC fusion vaccine anti-tumor effects . Methods: The NOD / SCID mice were divided into ABCDEF6 group, which BCDEF5 group rebuild human immune system then CDEF4 group Construction A498 (kidney cancer) tumor-bearing model in skin. CDE three groups were divided given the CD, GM-CSF modified fused cells, GM-CSF-B7.1 modified fused cells for treat tumor models, followed by monitoring changes in immune function between the groups and to the size of the tumor models. Results: The Mice in immunized construction group were detect human immune cells: CD3, CD45, CD56, and the numbers of human immune cell increase with time prolonged, peaked in the eighth week, then gradually decline. The number of immune cells in immune reconstitution group were descendants than immune reconstitution group .but after applying a DC fusion vaccine therapy, the number of human immune cells increased, and DC fusion vaccine significantly reduced the size of the kidney tumor tissue. Conclusion: Determine the DC fusion vaccine with enhanced immunity, and has a strong anti-tumor effect. Prove specific modifications of fusion vaccine has significantly improved of anti-tumor effect.
