Hedgehog信号通路在佐剂性关节炎大鼠关节软骨中的表达及其意义
Expression of hedgehog signaling pathway in articular cartilage of adjuvant-induced arthritis in rats and its pathologic significance
目的:研究Hedgehog (Hh)信号通路在佐剂性关节炎(AIA)大鼠关节软骨中的表达及其意义。方法:采用弗氏完全佐剂诱导大鼠AIA 模型,采用免疫组化和实时荧光定量PCR观察Hh信号通路重要分子Shh,Ptch1,Gli1在AIA大鼠关节软骨中的表达情况,并与AIA病理指征(足肿胀、关节病理学评分)进行相关性分析。结果:在成功复制AIA大鼠模型的基础上,发现AIA大鼠关节软骨中Shh,Ptch1,Gli1的蛋白和基因表达水平均明显高于正常对照组,并且Shh,Ptch1,Gli1 mRNA的表达水平与AIA大鼠的继发性足肿胀和膝关节病理学评分具有明显的正相关。结论:Hh信号通路在AIA大鼠关节软骨中异常活化,并密切参与AIA的发病过程。
Objective: We aimed to study the expression of hedgehog (Hh) signaling pathway in articular cartilage of adjuvant-induced arthritis (AIA) in rats and its pathologic significance. Methods: AIA in Rats was induced by injection of Complete Freund's adjuvant. The protein and mRNA levels of Hh pathway related genes (Shh, Ptch1 and Gli1) in articular cartilage were assayed by immunohistochemistry and real-time quantitative PCR respectively. In addition, we analyzed the possible relationships between Shh, Ptch1 and Gli1 mRNA levels and clinicopathologic parameters of AIA rats including secondary paw swelling and histological scores of knee joint damage. Results: We successfully established the AIA rat model. The protein and mRNA levels of Shh, Ptch1, and Gli1 showed higher levels in articular cartilage of AIA rats compared with the sham rats, and the elevated levels of Shh, Ptch1, and Gli1 mRNA were indeed positively associated with the secondary paw swelling and histological scores of knee joint damage. Conclusion: Hh signaling pathway was over-activated in articular cartilage of AIA rats, and the elevated Hh signal was apparently associated with AIA disease severity.
