Notch信号途径在无脊椎动物和脊椎动物中广泛存在且进化上十分保守的信号传递机制。它介导相邻细胞之间通讯,从而扩大并固化细胞间的分子差异,最终决定细胞命运,影响器官形成和形态发生。同时在肾脏发育过程中也发挥着及其重要的作用。新近发现它亦与肾脏疾病的发生发展关系密切。在正常发育成熟的肾单位中,Notch 信号途径的各个分子均为低表达。然而在肾小球疾病,急性肾损伤,肾小管间质纤维化及肾脏肿瘤中均可见Notch信号通路的异常活化,予以Notch 信号抑制剂后能有效减轻肾脏损伤。本文结合最新进展就Notch信号途径与肾脏疾病进行初步的探讨。?????
Notch signaling pathway exists widely in invertebrates and vertebrates, an evolutionary conserved cell-cell communication mechanism. It mediates communication between adjacent cells ,thus expands and solidifys intercellular molecular differences to decide cell fate and affect organ formation and morphogenesis. At the same time it plays an important role in the renal development,too. The recent findings verify that it also closely associated with the occurrence and development of kidney disease . In the normal mature nephron, almost of the molecules of notch signaling pathway are lower expression. However, a number of studies have confirmed notch signaling pathway is abnormal activation in glomerular disease, acute kidney injury, renal tubular interstitial fibrosis and kidney tumors, and its inhibitors can effectively reduce the kidney damage. This paper, combining the latest progress, aims to discuss Notch signaling and kidney disease.
