从晒盐场盐池周边散落的风干野生植被表面分离筛选到1株极端嗜盐菌株CCHA,通过形态鉴定及5.8S rDNA序列分析等方法对菌株进行鉴定,同时对其生物学特性、耐盐性及金属耐受性进行了分析。5.8S rDNA基因序列分析显示菌株CCHA与 21株己报道的曲霉属菌株同源性为55.9 %-100 %,表明该菌株为曲霉菌(Aspergillus sp.)。其生长温度为25 ℃- 45 ℃;生长pH为2.5-12.0;其最适培养条件为35 ℃,pH 5.0-8.0,盐浓度为5 %。常温下,该菌株能在NaCl 浓度高达500.0 g/ L的PDA培养基中生存;35 ℃下在含NaCl 310.0 g/ L的PDA培养基中存活60 d之久。其对Cu2+有较强的耐受性。另外该菌株对水稻、大豆、烟草和黄瓜没有潜在致病性。菌株CCHA是一株极端嗜盐且适应温度及pH范围较广的曲霉属菌株,具有重要的潜在应用价值。

We isolated and identified an extreme halophilic strain CCHA from the surface of wild vegetation around soalar saltern, and identified by the morphological characters and 5.8S rDNA sequence analysis, at the same time, their biological characteristics, salt-resistance and metal-tolerance was analyzed. 5.8S rDNA sequence analysis showed that the homology of CCHA and 21 Aspergillus strains reported was 55.9%-100 %. This indicates that CCHA belongs to Aspergillus sp.. Strain CCHA grew at temperature from 25 ℃ to 45 ℃; Growth pH range was from 2.5 to 12.0; Temperature 35 ℃, pH from 5.0 to 8.0, and salt concentration 5% was the optimum culture condition. At room temperature, the strain could survive on PDA up to 500.0 g / L NaCl; At 35 ℃, It could survive two months in PDA containing 310.0 g / L NaCl. It had higher tolerance to Cu2+. In addition, it has no potential pathogenicity to rice、soybean、tobacco and cucumber. CCHA strain was identified as Aspergillus sp. with extreme halophilic characteristics and adapt to wide range of temperature and pH, being important potential application value.
