目的:研究α-MEM、DMEM-LG、RPMI1640三种培养基对人牙周韧带细胞(human periodontal ligament cells, hPDLCs)生物学行为的影响,为体外培养人牙周韧带细胞寻找合适的培养基。方法:采用改良组织块培养法,分别用α-MEM、DMEM-LG、RPMI1640三种培养基对hPDLCs进行体外培养,比较三组细胞在游出成功率、增殖及矿化等方面的差异。结果:α-MEM促进hPDLCs增值作用最强。α-MEM组培养的细胞表达的碱性磷酸酶(Alkaline phosphatase,ALP)含量最高,与其它两组间有显著差异。DMEM-LG组矿化结节形成个数最多且体积大。结论:α-MEM促进hPDLCs增殖,DMEM-LG促进hPDLCs分化。应根据实验要求选择合适培养基。

Objective: To observe the effects of three different culture medium: α-MEM、DMEM-LG and RPMI1640 on the biological behavior of human periodontal ligament cells(hPDLCs), and provide the best culture medium for cultured hPDLCs in vitro. Methods: hPDLCs were cultured in α-MEM、DMEM-LG and RPMI1640 respectively in vitro with the improved tissue block culture method. The successful cell migration rate, proliferation, mineralized nodules of three groups cells were defected and compared. Results: α-MEM significantly increased hPDLCs proliferation. In addition,α-MEM strongly induced alkaline phosphatase activity. But DMEM-LG group exhibited the most mineralized nodules with biggest size. Conclusion: α-MEM increased hPDLCs proliferation. DMEM-LG promoted the differentiation of hPDLCs. It could be ascertained to choose appropriate culture medium according to the demand of different experiments.
