电针对大鼠脊髓损伤后notch1、ps1表达的实验研究

目的:探讨电针治疗对大鼠脊髓损伤后的保护机理。方法:大鼠随机分为模型组、空白组、药物组、电针组,用改良Allen‘s打击法制成脊髓损伤模型,分别于损伤后6h、1d、7d取损伤局部脊髓组织,提取总RNA,经RT-PCR反应,测定损伤后脊髓组织内Notch1、ps1 mRNA的表达。结果:脊髓损伤后,电针组和药物组均能显著降低脊髓损伤6h、1d、7d后脊髓组织中Notch1、ps1 mRNA的表达。与模型组相比较,药物组和电针组Notch1、ps1 mRNA表达均降低,并都有显著性差异(P<0.05);药物组与电针组相比较,电针组Notch1、ps1 mRNA表达略高于药物组,但无显著性差(P>0.05)。结论:Notch信号通路,在细胞的生长、发育、分化及凋亡中起着十分重要的作用,Notch信号的激活可加速并不可逆地促进NSC向胶质细胞分化,电针治疗对大鼠脊髓损伤后Notch1、ps1表达具有明显的抑制作用。

Objective:To discuss the protective mechanism of electroacupuncture (EA) in the spinal cord injury(SCI). Methods: Rats were divided into four groups randomly:Model Group,Blank Group,Drug Group and EA Group. The SCI rat model was made by using a modified Allen’s method. We will take the spinal cord tissues on the six hour, the first day and the seven day after injury. The injured tissues of Model Group,Blank Group,Drug Group and EA Group were removed to determine the content of Notch1、ps1. Total RNA was extracted,through RT-PCR.. Results: After the SCI rat model be made suceessfully,the Notch1、ps1 mRNA content of injury local tissues were decreased significantly in EA Group and Drug Group on the 6h, 1 day and the 7 day after SCI. Compared with the model Group,statistical significance were found in the EA Group and Drug Group(P<0.05). The Notch1、ps1 mRNA content of injury local tissues were higher slightly in EA Group than the Drug Group and there was no statistical significance between the Drug Group and the EA Group(P>0.05). Conclusion:Notch signaling pathway plays an important role in cell growth, development, differentiation and apoptosis. The activation of Notch signaling can reversibly accelerate the NSC changing into glial cell.The electric acupuncture on spinal cord injury can inhibit the expression of Notch1, ps1 significantly.
