ORF B 6.4-7.0 kb of hypoviruse regulates viral dsRNA accumulation
低毒病毒是一类能够降低板栗疫病菌致病力的无衣壳正链RNA病毒,其中低毒病毒CHV1-Euro7的dsRNA在真菌细胞中的积累量较高,而低毒病毒CHV1-EP721在真菌细胞中的积累量则明显较低。本研究构建了含有低毒病毒CHV1-Euro7 基因组ORF B中6.4-7.0 kb区域的反式互补重组质粒0.6GpCPXHY2和含有此区域但在其ATG三联体前加上两个TAA终止密码子的的重组质粒pCPTTP2,并分别用于转化携带CHV1-EP721病毒的菌株EP721。通过对阳性转化株的dsRNA半定量分析,发现0.6GpCPXHY2转化株可提高CHV1-EP721病毒在宿主细胞的累积量,但翻译终止质粒pCPTTP2转化株则丧失了此功能,表明:1)病毒 ORF B中6.4-7.0 kb区域是病毒dsRNA积累的一个决定因子;2)该因子以蛋白的形式作用。本研究为进一步弄清低毒病毒dsRNA在细胞的积累机理打下了新的基础。
Hypoviruses are a group of unencapsidated, plus-stranded RNA viruses,which can attenuate the virulence of the chestnut blight fungus, Cryphonectria parasitica. Among these viruses, CHV1-Euro7 accumulates much more viral dsRNA in the host cells than CHV1-EP721 does. In this study, we constructed an in trans complementary plasmid 0.6GpCPXHY2 which contains a 0.6kb fragment between 6.4kb to 7.0 kb located in the ORF B of the CHV1-Euro7, and a similar plasmid, pCPTTP2, which contains an untranslatable form of the 0.6kb fragment by addition of double TAA terminator codons in front of the ATG triplet of the fragment. Then the two plasmids were introduced into the protoplasts of the EP721 separately. Transformants containing a normal form of the viral ORF B 6.4-7.0kb fragment increased the dsRNA level of CHV1-EP721 in the host, but transformants with the untranslatable form of the ORF B 6.4-7.0kb fragment failed to elevate the dsRNA accumulation level. These results demonstrate that 1) viral ORF B 6.4-7.0kb fragment is a determinant for viral dsRNA accumulation and 2) this determinant functions in form of protein. Our work lays a new foundation for elucidation of the mechanism of hypovirus dsRNA accumulation in the host cells.
