西瓜和甜瓜蔓枯病菌分子检测技术研究

由Didymella bryoniae引起的西瓜和甜瓜蔓枯病是最重要的一种世界性的西瓜和甜瓜土传病害之一,快速有效地检测该病害对及时防治病害有着重要意义。鉴于蔓枯病具有种传和土传等特点,本文在筛选出较好的PCR和real-time PCR引物的基础上,对接菌后的西瓜植株、西瓜和甜瓜种子、接菌后的土壤做了检测研究。结果表明,利用PCR和real-time PCR技术能在蔓枯病发病前即可检测是否感病,能用于检测市面上所售西瓜和甜瓜种子以及瓜地土壤是否带菌,为西瓜和甜瓜的蔓枯病防治提供参考。
Gummy stem blight, caused by Didymella bryoniae, is one of the most important worldwide soil-borne diseases of watermelon and melon, and to found rapid and efficient methods of disease detection has huge significance for timely prevention and control. Considering that it is a seed-borne and soil-borne disease, the article screened couples of primers that suitable for PCR and real-time PCR, then used them to detect plant tissues of watermelon that inoculated with disease, seeds of watermelon and melon, and soil that inoculated with disease. The results show that PCR and real-time PCR can detect whether plant infected or not before disease occurring, whether the watermelon and melon seeds on the market carrying diseases or not, and whether melon soils containing disease or not, so that providing reference for GSB prevention and control.
