大雪素叶片蛋白质提取方法的比较
omparsion on Two Methods of Proteins Isolation from Cymbidium tortisepalum
本实验比较了(TCA)/丙酮沉淀法和改良的Tris-HCl法提取大雪素叶片蛋白质的差异以及不同方法制备的样品对SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析的影响，制备的样品经SDS-PAGE分离，采用考马斯亮蓝显色。结果表明，改良的Tris-HCl法对大雪素蛋白质的样品制备有较好的效果，制备的大雪素样品适用于SDS-PAGE分析。
In this study, the two methods for protein extraction of trichloroacetic acid/acetone precipitation(TCA) and modified Tris-HCl extraction leaf were compared in Cymbidium tortisepalum, meanwhile, the two different methods of protein samples prepared for SDS-polyacrylamide gel electrophoresis(SDS-PAGE) was analysized. The extracts were separated by SDS-PAGE method and followed by Coomassie blue color. The results showed that the modified Tris-HCl extraction can be used for the sample preparation of the C. tortisepalum, and it is also suitable for SDS-PAGE analysis.
