IFB Reaction Synthesis of 2,3-dihydro-furan Derivatives
二氢呋喃类化合物是一类重要的杂环化合物,化学性质特殊,广泛应用于药物及化学中间体合成,其中含手性2,3-二氢呋喃结构单元的砌块在聚醚类抗生素、木脂素、核苷及多种手性药物的合成中起重要作用。本文分别从苯甲酸乙酯和金刚烷甲酸出发合成了β-羰基腈类化合物苯甲酰乙腈和金刚烷甲酰乙腈,将其与3-溴代丙酮酸乙酯进行IFB反应,得到4-氰基-2,3-二氢呋喃类衍生物。
ihydrofurans are an important class of heterocyclic compounds. For their special chemical properties, they have found wide applications in the synthesis of pharmaceuticals and chemical intermediates etc. Chiral 2,3-dihydrofurans are highly important building blocks for the synthesis of numerous natural products (e.g. polyether antibiotics, lignans, nucleoside) and some chiral pharmaceuticals.This work, synthesis of β-carbonyl nitriles benzoylacetonitrile and adamantane formyl acetonitrile was prepared with adamantane formic acid and ethyl ester, which was carried out with ethyl bromopyruvate to give 4-cyano furan-2,3-dihydro derivatives.
