目的:验证二甲双胍减轻小鼠体重的分子机制。方法:将小鼠分为普通饲料组(CON),高脂饲料组(HFD)及二甲双胍组(HFD+MET),喂养22周后取皮下及内脏脂肪检测相关基因的表达水平。结果:与HFD组相比,二甲双胍显著减轻了小鼠的体重及脂肪重量。并且,HFD+MET组特异性下调了脂肪细胞中胰岛素受体的表达水平。结论:二甲双胍可能是通过特异性下调脂肪细胞中胰岛素受体的表达,减少脂肪的合成进而达到减轻体重的效果。

In this paper, we investigated the molecular mechanism of metformin on weight loss in mice.Mice were randomly divided into CON group (standard diet), HFD group (high-fat diet), and HFD+MET (metformin) group. Mice in HFD+MET group showed lower body weight and white adipose tissue (WAT) mass in comparison to those in HFD group.Adipogenic genes were down-regulated after metformin treatment. Interestingly, metformin treatment down-regulated the protein level of insulin receptor (IR) whereas, up-regulated the protein level of IR in skeletal muscle.Our results indicated that metformin prevented weight gain through the specific down-regulation of insulin signaling pathway in WAT.
