目的 探讨胰岛素对SGC7901细胞极低密度脂蛋白受体(VLDLR)亚型表达的影响及其可能的生物学意义。 方法 在体外,用胰岛素与中低分化的胃腺癌细胞株SGC7901温育不同时间,采用Western Blotting技术检测VLDLR亚型蛋白表达的变化,Real-Time PCR技术检测VLDLR亚型mRNA水平变化。 结果 在一定时间范围内,胰岛素可使VLDLR I型表达显著下调,II型表达显著上调,且mRNA水平结果与蛋白水平结果相一致。 结论 胰岛素可能通过改变体内VLDLR亚型的差异性表达,在各种生理与病理条件发挥对糖代谢与脂代谢的协调控制作用。

Objective To examine the effect of insulin on the expression of Very low density lipoprotein receptor(VLDLR) subtypes of SGC7901 cells and its possible biological Significance. Methods In vitro, moderately or poorly-differentiated human gastric adenocarcinoma cell line SGC7901 was incubated with insulin for different time, then the expression of VLDLR subtypes in protein and RNA level were detected by Western Blotting and Real-Time PCR techniques, respectively. Results In certain time interval, insulin could down-regulate expression of type I VLDLR and up-regulate the expression of type II VLDLR in SGC7901 cell, both in protein and in RNA level. Conclusion Insulin could exert its regulating rule on maintaining the balance between glucose and lipid metabolism in vivo , by means of its effects on differential expression of VLDLR subtypes.
