柚皮苷对脂多糖诱导的beagle犬急性肺损伤的保护作用
Protective effects of naringin on lipopolysaccharide induced acute lung injury in beagle dogs
目的:研究柚皮苷对脂多糖诱导的beagle犬急性肺损伤的保护作用。方法:beagle犬18只,随机分为安慰剂组、LPS+安慰剂组、LPS+柚皮苷组。LPS+柚皮苷组按12.4 mg/kg的剂量给予柚皮苷胶囊,安慰剂组与LPS+安慰剂组给予空白胶囊壳。给药1 h后,LPS+安慰剂组与LPS+柚皮苷组气管滴注脂多糖4 mg/kg构建急性肺损伤模型。造模8 h后取血和支气管肺泡灌洗液及肺部组织,检测肺湿干比、痰液中总蛋白、Cl-、K+和Na+含量、肺组织中性粒细胞髓过氧化物酶(myeloperoxidase,MPO)和诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)的活性、支气管肺泡灌洗液和痰液中白介素-8(interleukin-8,IL-8)与肿瘤坏死因子(tumor necrosis factor-α,TNF-α)的含量,并采用HE染色观察肺部组织病理变化。结果:柚皮苷能显著降低肺组织的湿干比、iNOS和MPO活性,减轻肺部白细胞及中性粒细胞浸润、抑制LPS引起的TNF-α与IL-8分泌、减轻肺部组织炎性病理损伤与出血渗出。结论:柚皮苷预处理对LPS所致的急性肺损伤具有保护作用。
Objective: To observe the potential protective effects of naringin on the lung tissue in lipopolysaccharides (LPS)-induced acute lung injury (ALI) mice. Methods: Eighteen beagle dogs were randomly divided into normal control, LPS alone, and LPS+naringin groups. Dogs in LPS+naringin group were pretreatment with naringin at dose of 12.4 mg/kg, while in LPS and normal control groups were dealt with placebos. 1 h latter, LPS (4 mg/kg) or saline was instilled intratracheally followed in 8 h of modeling period. Results: Naringin significantly decreased LPS-induced influx of inflammatory cells, the contents of total protein and cytokines production such as TNF-α and IL-8 in BAL fluid, while significantly but only partially inhibited the activities of MPO and iNOS in lung tissue. Moreover, naringin markedly attenuated LPS-induced increases total sputum volume, as well as pulmonary edema. Conclusion:pre-oral administration of naringin reduces inflammatory responses, pulmonary edema, sputum synthesis, and damage of lung tissue in LPS-induced ALI beagle dogs.
