针灸疗法对EAMG大鼠血清中AchR-ab和IFN-γ表达水平的影响
Effect of acupuncture on AchR-ab and IFN-γ in serum of EAMG rat
目的:观察针灸疗法对实验性自身免疫性重症肌无力(EAMG)大鼠血清中乙酰胆碱受体抗体(AchR-ab)、干扰素-γ(IFN-γ)表达水平的影响,分析针灸治疗重症肌无力(MG)的作用机制。方法:采用AchRα1 129-145多肽片段免疫接种雌性Lewis大鼠,建立EAMG大鼠动物模型;随机选取建模成功的EAMG大鼠30只,分成针灸治疗组、药物对照组、模型对照组各10只,并将同期购进未建模的10只大鼠设为空白对照组;针灸治疗组予以"温阳补气"针法治疗,药物对照组予以溴吡斯的明灌胃治疗,模型对照组和空白对照组只进行与其他两组相同的抓取。酶联免疫吸附法(ELISA)检测大鼠血清中AchR-ab、IFN-γ的含量。结果:模型对照组大鼠血清中AchR-ab、IFN-γ表达水平,明显高于空白对照组,差异具有统计学意义;针灸治疗组、药物对照组大鼠血清中AchR-ab、IFN-γ表达水平,明显降低于模型对照组,差异具有统计学意义。结论:针灸可以降低EAMG大鼠血清中AchR-ab、IFN-γ的表达水平,达到治疗MG的作用。
Objective: Observation the influence of acupuncture treatment to the EAMG rats' serum AchR-ab and IFN-γ expression level, analyzes the mechanism of acupuncture treatment to MG. Method: Use AchRα1 129-145 peptide fragments to immunization Lewis female rats, for establish EAMG rat models. Selected 30 EAMG model rats, were randomly divided into acupuncture treatment group, drug control group, model control group, with 10 rats in each group. The unmodelled 10 rats which were purchased at the same period were set to blank control group. The acupuncture treatment group to be "warming yang and nourishing qi" acupuncture treatment, the drug control group to be the the pyridostigmine treatment, rats in model control group and blank control group were just captured by the same methods with other groups. The contents of AchR-ab and IFN-γ were tested by ELISA. Result: Model control group rats serum AchR-ab, IFN-γ expression level, is obviously higher than that of the blank control group, difference has statistical significance. The acupuncture treatment group and drug control group rats serum AchR-ab, IFN-γ expression level, decrease in model control group, the difference is statistically significant. Conclusion: The "warming yang and nourishing qi" acupuncture can obviously improve the ENMG rat myasthenia gravis symptoms, and reduce the EAMG rats' serum AchR-ab and IFN-γ expression level, thus giving full play to the role of MG treatment.
