目的:研究紫云英苷对体外培养成骨细胞MC3T3-E1 增殖、分化、矿化及OPG/RANK/RANKL的影响。方法:采用MC3T3-E1 成骨细胞株为体外实验模型,使用不同浓度紫云英苷干预MC3T3-E1 细胞,以MTT 法测定成骨细胞的增殖率,试剂盒比色法检测ALP活性观察不同浓度的紫云英苷溶液的促细胞分化作用,用茜素红染色法考察紫云英苷的促细胞钙化作用,ELISA 方法检测OPG 和RANKL 的表达水平,并计算OPG/RANKL 的比值。结果:与对照组相比,紫云英苷在(5-100μM)的浓度范围内能显著的促进MC3T3-E1 细胞增殖( P<0.05);紫云英苷低、中、高三个浓度(5、10、20μM)干预细胞10天能够显著提高ALP 的活性(P<0.05) ;紫云英苷低、中、高三个浓度(5、10、20μM)干预细胞28天能够显著增加MC3T3-E1细胞的矿化结节。同时,紫云英苷在20μM浓度水平,可明显抑制RANKL 的表达(P<0.05),促进OPG的表达,上调OPG/RANKL的比值。结论:紫云英苷可通过促进MC3T3-E1成骨细胞增殖与分化成熟,并上调OPG/RANKL的比值,间接抑制破骨细胞的分化和成熟。提示紫云英苷具有潜在的抗骨质疏松作用。

Objective:This study aimed to evaluate effects of Astragalin on the proliferation, differentiation, calcification and OPG/RANK/RANKL axis in mouse osteoblastic MC3T3-E1 cells.?Methods:MTT assay was applied in the evaluation of MC3T3-E1 cells proliferation. Differentiating actions of Astragalin with different concentrations in cells were investigated through the examination of alkaline phosphatase (ALP) activities by colorimetric method. And bone nodule formation was dectcted via Alizarin red S staining.The proteins expression of OPG and RANKL of MC3T3-E1 was assayed was assayed using ELISA kit. Results:The effective concentration of Astragalin on MC3T3-E1 proliferation was 5 -100μM. In additional, the ALP activity of MC3T3-E1 could be increased by quercetin (5, 10 and 20μM). And The Astragalin at the concentration (5, 10 and 20μM) remarkably increased the bone nodule formation of cells.The Astragalin could also obviously decrease (P<0.05) the expression of the RANKL, and increase the expression of OPG obviously (P<0.05). Ultimately, the Astragalin upgraded the ratio of OPG/RANKL (P<0.05).Conclusion:Astragalin could promote the proliferation and differentiation of osteoblasts, as well as enhance the ratio of OPG to RANKL, which indirectly inhibit bone resorption process. The results indicated that Astragalin may be an effectve treatment for osteoporosis that acts, in part, via a decrease in bone resorption and an increase in bone formation.
