梨CYP707A基因特征及其在花芽休眠期间的表达分析

YP707A基因编码ABA-8'羟化酶,ABA分解途径上的关键酶,本文为探讨梨CYP707A基因与花芽休眠的关系,选用'黄花梨'(Pyrus pyrifolia.'Huanghua')一年生饱满花芽,分离出梨PpCYP707A1基因(Genbank登录号:KP057206),研究'黄花梨'PpCYP707A1在花芽休眠期间的表达情况。生物信息学分析结果表明,PpCYP707A1基因CDS序列全长1359bp,编码452个氨基酸,蛋白理论分子质量为51.32KDa,为不稳定的亲水性蛋白,具有1个PLN02196保守结构域,属于P450超基因家族。QRT-PCR分析结果显示PpCYP707A1在黄花梨花芽中的年际表达较为稳定,出现了多次表达高峰,表明PpCYP707A1参与了'黄花梨'芽休眠期间对环境胁迫的应答,以及芽休眠解除的调控。
he CYP707A has been identified as ABA 8'-hydroxylase, which is the pivotal enzyme in the oxidative catabolism of ABA. In order to understand the relationship between CYP707A gene of pear and flower buds dormancy, the PpCYP707A1 (GenBank Accession Number: KP057206) was obtained from annual full flower buds of Pyrus pyrifolia.'Huanghua'. Then, the gene expression patterns in flower buds of pear cultivars 'huanghua' was analyzed during the period of dormancy by QRT-PCR. The results indicated that the full-length CDS of PpCYP707A1 was 1359bp bp which encoded 452 amino acid residues with a putative molecular mass of 51.32 kD. It was predicted that PpCYP707A1 protein of pear cultivars 'huanghua' was hydrophilic protein and contained a PLN02196 putative conserve domains of P450 super family. QRT-PCR analysis showed that the expression patterns of PpCYP707A1 gene in two years were similar with many peaks during the period of dormancy. Results suggested that PpCYP707A1 might involve in the response of stress and the regulation of bud dormancy release of 'Huanghua'
