产生物碱抗氧化"金花"菌的筛选及沙棘叶发酵应用
Screening of Alkaloid-Producing "Golden Flower" Fungi with Antioxidant Activity and the Application in Hippophae rhamnoides Leaves Fermentation
从茯茶中筛选高产生物碱且抗氧化活性强的"金花"菌,制备发酵沙棘叶茶(FHRL)。通过比较发酵前后营养成分(总酚、总黄酮、游离氨基酸、可溶性糖、总蛋白、咖啡因和neoechinulin A)及抗氧化活性(总还原力、DPPH 清除率、 OH清除率)差异,评价"金花"菌对沙棘叶的影响,进行模拟消化实验观察FHRL的生物可及性。所得菌株Eurotium amstelodami JH-3的粗吲哚生物碱和neoechinulin A产量分别为28.64 μg/g、0.80 mg/g;发酵水提取液的总还原力、DPPH 清除率、 OH清除率为0.68 mmol/L、51.89%、35.85%;发酵醇提取液的总还原力、DPPH 清除率、 OH清除率为2.06 mmol/L、96.82%、29.56%。用其制备FHRL,总黄酮含量提高了34.86%,neoechinulin A含量达到42.92 mg/L,总酚变化不显著,可溶性糖、氨基酸下降,咖啡因含量显著下降。总还原力和DPPH 清除率分别提高了32.95%和22.42%, OH清除率显著下降。对FHRL进行模拟消化实验,发现除可溶性糖和氨基酸以外,各类营养成分均呈现下降趋势。消化后总还原力和DPPH 清除率分别下降60.59%和21.09%, OH清除率显著提高。本研究为传统微生物资源和沙棘叶的开发利用了提供参考。
high alkaloid-producing fungus with strong antioxidant activity was screened from Fu tea and used to prepare fermented Hippophae rhumnoides leaf (FHRL).The effect of "Golden Flower" fungi on Hippophae rhumnoides leaves was evaluated by comparing the differences in nutrient composition (total phenols, total flavonoids, free amino acids, soluble sugars, total proteins, caffeine and neoechinulin A) and antioxidant activity (total reducing power, DPPHo scavenging ability, oOH scavenging ability) before and after the fermentation, and then simulated digestion experiments were carried out to observe the bioaccessibility of FHRL. The yields of crude indole alkaloids and neoechinulin A of Eurotium amstelodami JH-3 were 28.64 μg/g and 0.80 mg/g, respectively, and the total reducing power, DPPHo scavenging rate, and oOH scavenging rate of the fermented aqueous extract were 0.68 mmol/L, 51.89%, and 35.85%, respectively. The total reducing power, DPPHo scavenging rate, and oOH scavenging rate of the fermented methanol extract were 2.06 mmol/L, 96.82%, and 29.56%. Preparation of FHRL with this strain resulted in a 34.86% increase in total flavonoids, 42.92 mg/L of neoechinulin A, insignificant changes in total phenols, decreases in soluble sugars and amino acids, and a significant decrease in caffeine content. Total reducing power and DPPHo scavenging rate increased by 32.95% and 22.42%, respectively, and oOH scavenging rate decreased significantly. Simulated digestion experiments on FHRL revealed a decreasing trend in all types of nutrients except soluble sugars and amino acids. The total reducing power and DPPHo scavenging rate decreased by 60.59% and 21.09%, respectively, and the oOH scavenging rate increased significantly after digestion. This study provides a reference for the development and utilization of traditional microbial resources and Hippophae rhumnoides leaves.
