瘦素与雌激素对间充质干细胞的协同作用--绝经后骨质疏松症发病机制探讨
Synergistic effect of leptin and estrogen on mesenchymal stem cells-Investigation of pathogenesis of postmenopausal osteoporosis
目的:研究瘦素对骨髓间充质干细胞(MSCs)自我更新能力的作用及雌激素影响下瘦素作用的变化。方法:二月龄雌性SD大鼠按体重随机分层为去势组和假手术组,术后14周检测大鼠股骨骨密度及血清瘦素及雌激素水平,同时以成骨前体细胞MSCs为模型,CCK8法检测瘦素,雌二醇及两者共同添加在体外对MSCs增殖的影响,采用定量PCR检测细胞周期相关基因表达水平,western blot检测细胞Akt磷酸化水平。结果:去势14周后股骨头及股骨干骨密度均较假手术组明显下降,瘦素与雌二醇的交互作用对骨密度有保护作用;与试剂对照组相比,在干预第6天瘦素(100ng/ml)能明显增加MSCs增殖水平,在添加雌激素后对MSCs的增殖水平增加更显著;雌激素与瘦素均可促进cyclinD1表达,瘦素还能显著促进cyclinD2、PCNA表达;WB检测发现瘦素、雌激素可促进Akt磷酸化,两者联用对Akt磷酸化有时间依赖性的协同作用。结论:瘦素在体外可促进MSCs增殖,雌激素和瘦素对MSCs自我更新能力的协同作用可能是通过Akt信号途径实现。
Objective: To study the influence and mechanism of leptin, estradiol, leptin and estradiol together on self-renewal capacity in bone marrow mesenchymal stem cells (BMSCs). Method: We stratifiedly randomized two-month-old female SD rats into bilateral ovariectomy (OVX) and sham group according to body weight. 14 weeks after the surgery, bone mineral density (BMD) were measured by dual energy X ray absorptiometry (DXA), seurm leptin and estradiol lever were examinated by Enzyme linked immunosorbent assay (ELISA) and radioimmunoassay respectively. We introduced CCK-8 to figure out the effect of leptin, estradiol, leptin plus estradiol on BMSCs proliferation, adopted qPCR to evaluate mRNA expression of cell cycle related gene, and introduced western blot to examine lever of phosphorylation and protein expression of Akt. Result: BMD of femoral head in OVX group was significantly reduced, and so dose the BMD of femoral shaft. The interaction between estrogen and leptin might had protective effect on mineral density of femoral head. After intervetion for 6 days, leptin but not estradiol can obviously promote proliferation of BMSCs. Leptin plus estradiol had synergistic effect to encourage BMSCs proliferation. mRNA expression of Akt downstream moleculors (cyclinD1, cyclinD2 and PCNA) was elevated in leptin treatment group. Further western blot detection found that both estrodiol and leptin promoting Akt phosphorylation in BMSCs in every time point And the lever of Akt phosphorylation demonstrated time dependent effect under adiministration of both two hormones. Conclusion: Leptin can stimulate proliferarion of BMSCs in vitro. Leptin and estradiol have synergitic effect in promoting self-renewal capacity of BMSCs through Akt signal pathway.
