目的:探讨干燥综合征外分泌腺免疫炎性损伤的分子机制及半夏芩连汤的干预作用。方法:将24只雌性NOD小鼠随机分为模型组、阳性药组、中药双倍剂量组、中药常规剂量组,以雌性Balb/c小鼠设为空白对照组,每组6 只。空白对照组及模型组予去离子水0.1ml/10god灌胃,阳性组予雷公藤多甙片10mg/kgod灌胃,中药双倍剂量组、常规剂量组每日予半夏芩连汤按含生药120g/kg、60g/kg灌胃。连续给药12周后,留取血清、双侧腮腺、颌下腺标本。采用RT-PCR检测AMPKα1、TNF-αmRNA转录水平,Western Blot及免疫组化技术检测AMPKα1蛋白表达水平, ELISA法检测TNF-α蛋白表达水平。结果:①RT-PCR检测结果:与正常组比较,模型组小鼠腺体AMPKα1 mRNA转录水平显著降低,TNF-α mRNA转录水平显著升高,差异具有统计学意义(P<0.01)。与模型组比较,阳性药组、中药各组AMPKα1 mRNA转录水平显著升高,TNF-α mRNA转录水平显著降低,差异具有统计学意义(P<0.01)。②Western Blot检测结果:与正常组比较,模型组小鼠腺体组织AMPKα1表达降低(P<0.05)。与模型组比较,阳性药组、中药各组AMPKα1表达升高,差异具有统计学意义(P<0.05)。③免疫组化检测结果:与正常组比较,模型组小鼠腺体组织AMPKα1蛋白表达减少(P<0.05)。与模型组比较,阳性药组、中药各组AMPKα1蛋白表达无统计学意义(P>0.05)。④ELISA检测结果:与正常组比较,模型组小鼠血清TNF-α蛋白表达水平明显增高(P<0.01)。与模型组比较,阳性药组、中药各组TNF-α蛋白表达水平显著降低(P<0.01)。结论:半夏芩连汤通过调控AMPK信号通路,下调促炎因子TNF-α表达水平,抑制SS外分泌腺免疫炎性病理损伤。

Objective: To investigate the molecular mechanisms of exocrine immune inflammatory injury of Sj?gren Syndrome and the intervention effect of Ban Xia Qin Lian decoction. Methods: Randomly divided the 24 female NOD mice into model group, positive drug group, double dose TCM group and Conventional dose TCM group, with 6 female Balb/c mice as normal control group. The mice of control group and model group were gavaged with deionized water(0.1ml / 10g o d), the positive group was intervened with Tripterygium (10mg / kg o d), the mice in TCM group were gavaged with chinese herbal compound Ban Xia Qin Lian decoction (Containing crude drug 120 g/kgod and 60g/kgod). The intervention and treatment time was 12 weeks. Then collect Serum, bilateral parotids and submandibular glands. samples .To detect AMPKα1、TNF-α mRNA transcriptional level by RT-PCR,;To assess AMPKα1 protein expression levels by western Blot and Immunohistochemical method;To observeTNF-α protein expression levels by ELISA method. Results: RT-PCR test results: compared with the normal group, the AMPKα1 mRNA transcription level was significantly reduced in the model group, TNF-α mRNA transcription level was significantly increased, the difference was statistically significant (P < 0.01). Compared with the model group, AMPKα1 mRNA transcription level was significantly elevated in the positive drug group and TCM groups,TNF-αmRNA transcription level was significantly reduced in the positive drug group and TCM groups, the difference was statistically significant (P < 0.01). Western Blot test results: compared with the normal group, the protein expression level of AMPKα1was decreased in the model group (P < 0.05). Compared with model group, the protein expression level of AMPKα1 was increased in positive drug group and TCM groups (P < 0.05). Immunohistochemical detection results: compared with the normal group, the protein expression level of AMPKα1 was decreased in the model group (P < 0.05). Compared with the model group, the protein expression level of AMPKα1 was not significant in the positive drug group and theTCM groups (P > 0.05). The ELISA test results: compared with the normal group, the protein expression level of TNF-αincreased significantly in the model group (P<0.01). Compared with the model group, the protein expression level of TNF- αwas significantly decreased in the positive drug group and the TCM groups (P<0.01).
