补阳还五汤对高血压大鼠肾脏KKS信号通路表达影响

目的:探讨补阳还五汤对高血压前期Dahl盐敏感大鼠肾脏KKS信号通路的影响。方法:取Dahl盐敏感大鼠20只,随机分为模型组和给药组。盐抵抗大鼠10只作为正常组。高盐喂养使血压逐步升高,在盐敏感大鼠血压值升至120-139/80-89mmHg时,停止高盐喂养,治疗组进行补阳还五汤灌胃治疗。5周后治疗结束,取大鼠左肾组织,荧光定量PCR法检测大鼠KKS信号通路相关基因表达;Elisa检测大鼠左肾组织NO含量。结果:与正常组相比,模型组Kng基因表达显著升高(P<0.05),KLK基因表达显著降低(P<0.05),NO含量显著降低(P<0.05);与模型组相比,中药组Kng基因表达显著降低(P<0.05),KLK、B2R基因表达显著升高(P<0.05),NO含量显著升高(P<0.05)。结论:补阳还五汤对高血压前期大鼠肾脏的保护作用与KKS信号通路密切相关。

Objective: To investigate the effect of Buyang Huanwu decoction on KKS signal pathway in the kidney of Dahl salt sensitive rats in pre-hypertension. Method: Dahl salt sensitive rats 20 were randomly divided into model group and administration group. Salt resistant rats 10 as normal group. Through high salt feeding to raise blood pressure, blood pressure in rats rose to 120-139/80-89mmHg, stop the high salt feeding, the medicine group of Buyang Huanwu decoction treatment. After 5 weeks, the left kidney tissue of rats was removed, and the KKS signal pathway related genes were detected by fluorescence quantitative PCR. Elisa was used to detect the content of NO. Result: Compared with the normal group, model group, Kng gene expression increased significantly (P<0.05), the expression of KLK and B2R were significantly reduced (P<0.05), decreased the content of NO (P<0.05); compared with the model group, the expression of Kng gene in Chinese medicine group decreased significantly (P<0.05), KLK, B2R gene expression increased significantly (P<0.05), the content of NO increased significantly (P<0.05).Conclusion: The protective effect of Buyang Huanwu decoction on the kidney in the pre-hypertension is closely related to the KKS signaling pathway.
