苯醌诱导小鼠骨髓lin-细胞毒性作用的研究
Research on Cytotoxicity of Mouse Bone Marrow Lin- Cells Induced by Benzoquinone
目的:初步研究苯醌(1,4-benzoquinone,BQ)对小鼠骨髓lin-细胞的毒性作用和诱导凋亡的作用机制。方法:用MACS(磁性细胞分选技术)从ICR小鼠股骨骨髓特异性分选lin-细胞,以1μM、5μM、10μM的1,4-对苯醌对lin-细胞染毒,并设阴性对照,每组设3个平行。24h后以MTT法测定细胞活力,流式细胞仪检测细胞凋亡情况,同时用实时荧光定量PCR检测凋亡Caspase-3基因的表达。结果:与对照组比较,5μM和10μM组细胞的相对增值率降低(P<0.05);暴露于1μM苯醌组细胞的凋亡诱导率显著升高(P<0.05);Caspase-3在1μM组的表达上调,在5μM和10μM组的表达下调。结论:苯醌对骨髓lin-细胞增殖生长的最大无作用浓度为1μM,此剂量的苯醌可明显诱导骨髓lin-细胞的凋亡,而超过此浓度,则细胞不表现凋亡的病理性防御。
Objective To explore the cytotoxicity of lin- cells from mouse bone marrow induced by 1,4-benzoquinone (BQ) and the mechanism of apoptosis. Methods Lin- cells were acquired from mouse bone marrow of ICR mouse's femurs and tibias by magnetic activated cell sorting (MACS). The cells with the concentration of 1×106/ml were exposed to BQ with the concentrations of 0μM, 1μM, 5μM and 10μM for 24h. Cell proliferation inhibition was detected by MTT assay. Apoptosis was determined by double stained assay with Annexin-Ⅴand PI. Caspase-3 mRNA level was determined by real-time PCR. Results Compared with the control group, the cell relative growth rate (RGR) decreased in 5μM and 10μM BQ treated group(P<0.05);the apoptosis rate of lin- cells treated with 1μM BQ increased significantly(P<0.05);an increased level of caspase-3 was observed in 1μM BQ treated group while the expression of caspase-3 appeared to be suppressed in 5μM and 10μM BQ treated group relative to control group. Conclusion The maximal non-effect concentration of BQ to mouse bone marrow lin- cells was 1μM, significant apoptosis in mouse bone marrow lin- cells was induced by BQ at the maximal non-effect concentration but not at higher concentrations.
