Modulation of 17β-estradiol on neuroexcitability by HCN channel in Ah-type baroreceptor neurons.
目的:研究17β-雌二醇 (17β-E2) 对雌鼠中兴奋阈值低、性别特异性分布的髓鞘化Ah型压力感受器神经元兴奋性的影响,并探讨其可能的作用机制。方法:迷走神经传入神经元的分离,主动脉减压神经标记,膜片钳电生理电生理技术,内脏感觉神经元的鉴别。结果:17β-E2潜在地激活了G蛋白偶联受体,可同步协同地调节HCN介导的AHPfast、ADPlate及RMP与阈电位。结论:17β-E2可经HCN增强Ah神经元的兴奋性。
Objective: To study the effects of 17β-estradiol on neuroexcitability in Ah-type baroreceptor neurons and to explore the involved mechanisms. Methods: Isolation of vagal afferent neurons, Aortic depressor nerve labeling, Electrophysiological techniques,Identification of neuronal cell types and data analysis. Results: Potentiated action of 17β-E2 on neuroexcitability is due to G-protein coupled receptor activation,HCN-mediated alternations in AHPfast, ADPlate, and RMP/threshold were synchronously and synergistically modulated by 17β-E2. Conclusion: The 17β-E2-mediated potentiation is confirmed in Ah-type baroreceptor neurons.
