杨树蛋白酶抑制剂基因在拟南芥中的过量表达及对小菜蛾的抑制作用
Overexpression of Populus proteinase inhibitor genes in Arabidopsis and inhibiting effects on the diamondback moth
蛋白酶抑制剂是植物自然防御体系的重要组成部分,也是植物抗虫基因工程中的重要候选基因。本试验通过农杆菌介导,将从杨树中分离的两个蛋白酶抑制剂基因(PtdKTI和PtdLPI基因)导入到拟南芥中,使其在拟南芥中获得高效表达。在对转基因拟南芥进行筛选后,我们选择目的基因表达量最高的纯合体植株饲喂小菜蛾,抗虫试验结果表明以转基因拟南芥饲食的小菜蛾在成活率、结茧率、化蛾率等方面都明显低于对照组,证明杨树来源的两个基因可使拟南芥对小菜蛾的抗性明显增强,从而为培育转基因抗虫植物新品种提供了新的途径。
Proteinase inhibitors are important for plant natural defense system, and they are also important candidate genes in genetic engineering for plant insect-resistance. By Agrobacterium mediated transformation, we transferred two proteinase inhibitor genes from Populus (PttKTI and PttLPI) into Arabidopsis for constitutive expression. After screening transgenic Arabidopsis, we choose those with highest expression level to feed the diamondback moth. Bioassay results indicated that insects fed on transgenic Arabidopsis had lower survival rate, cocooning rate, and moth rate than those fed on the control plants. This indicated that these two poplar protease genes could enhance the insect resistance of Arabidopsis, so that offerd a new way to breed genetically modified plants for better insect resistance.
