目的 研究微波固相合成法制备的低聚半乳糖(GOS)和低聚木糖(XOS)对高脂日粮诱导肥胖小鼠肠道氧化还原状态及菌群结构的影响。方法 32只昆明种小鼠随机分为4组:正常组、高脂组(18%猪油)、高脂+GOS组(高脂饲料中添加0.15%GOS)、高脂+XOS组(高脂饲料中添加0.15%XOS)。饲喂12周后测定小鼠的十二指肠、空肠、回肠、盲肠、结肠的抗氧化能力和氧化还原水平,并观察小鼠粪便中肠道细菌含量变化情况。结果 高脂日粮能够导致小鼠肠道氧化应激,降低肠道抗氧化能力,造成肠道有益菌和优势菌数量显著下降(P<0.05),大肠杆菌、粪肠球菌等腐败菌数量显著升高(P<0.05)。GOS和XOS处理可显著提高肠道粘膜T-AOC和CAT活力及GSH/GSSG水平,降低肠道的MDA水平,显著提高肠道乳杆菌、双歧杆菌数量,降低大肠杆菌、粪肠球菌数量(P<0.05)。结论 微波固相法合成的低聚糖能够改善高脂日粮造成的肠道氧化还原状态和菌群失衡。

Objective To study the antioxidative and prebiotic effects of oligosaccharides (GOS and XOS) produced by microwave assisted solid phase synthesis on mice fed high-fat diets. Method Thirty-two mice were randomly divided into 4 groups: control group, high-fat group (18%lard), high fat plus 0.15% GOS group, high fat plus 0.15% XOS group. The antioxidant ability and redox state of duodenum, jejunum, ileum, caecum and colon were detected and the fecal bacteria were also analyzed at the end of 12th week. Result High-fat diets not only induced oxidative stress and decreased the antioxidant capacity in mice, but also improved the spoilage bacteria percentage and inhibited beneficial bacteria growth in intestinal tract. Addition of GOS and XOS in high-fat diets could enhance the activities of T-AOC and CAT, improve the level of GSH/GSSG and decrease MDA content significantly (P<0.05). Moreover, GOS and XOS increased Lactobacillu, Bifidobacteria and other predominant bacteria, and reduced E. faecalis, E.coli and other putrefying bacteria dramatically(P<0.05). Conclusion: Oligosaccharides could accommodate the balance of redox state and microorganism composition of intestinal tract in mice fed high-fat diets.
