NLRP3炎症小体是由NOD样受体家族成员NLRP3蛋白与接头蛋白ASC及caspase-1形成的一个多蛋白复合物,其活化后能促进IL-1β、IL-18和IL-33等多种炎性细胞因子的剪切、成熟与分泌,因而在炎症发生过程中起关键作用。NLRP3炎症小体不仅能识别病毒或者细菌产生的病原微生物相关模式分子,还能被多种机体内外的"危险信号"所活化,其中包括多种代谢产物,如高血糖、饱和脂肪酸、胆固醇结晶、尿酸结晶等。近年来,NLRP3炎症小体的活化机制受到了极大的关注,越来越多的研究证实NLRP3炎症小体异常活化参与肠炎、肿瘤、痛风、神经退行性疾病、糖尿病和肥胖症等多种炎症性疾病的发生过程,本文将从NLRP3炎症小体如何调控这些疾病的发生发展方面进行综述。

he NLRP3 inflammasome is a protein complex composed of NLRP3 protein, the adaptor protein ASC and pro-caspase-1 protein, its activation can promote the secretion and maturation of IL-1β, IL-18 and IL-33, thereby it plays a key role in the inflammatory responses. The NLRP3 inflammasome not only can recognize viral or bacterial pathogens associated molecular patterns, but also can be activated by a variety of "danger signals" inside and outside of the body, including a variety of metabolites, such as hyperglycemia, unsaturated fatty acid, cholesterol crystal and uric acid crystals etc.. Recent years, the activation mechanisms of the NLRP3 inflammasome have been caused a great deal of attention, more and more researches have confirmed that the abnormal activation of NLRP3 inflammasome takes part in inflammatory bowel disease, cancer, gout, neurodegenerative diseases, diabetes, obesity and other inflammatory diseases. This review will provide an overview of how the NLRP3 inflammasome controls the occurrence and development of these diseases.
