柑桔木虱体内细菌多样性的DGGE和RFLP分析

对野外采集的柑桔黄龙病的传播媒介昆虫柑桔木虱体内的微生物群落的多样性进行了研究。利用基于 16S rRNA基因作为分子标记的变性梯度凝胶电泳(denaturing gradient gel electrophoresis, DGGE)的方法和基于16S rRNA基因的RFLP指纹图谱法进行分析。通过RFLP方法获得的细菌类群包括Pseudomonadaceae, Enterobacteriaceae, Xanthomonadaceae, Burkholderiaceae,Rickettsiaceae,Rhizobiaceae等6个科的微生物。另外还有属于柑桔木虱的合胞内共生菌(Syncytium endosymbiont) 和次级共生菌(Secondary symbiont)。DGGE法获得的结果与RFLP稍有不同,包含Proteobacteria 4个科的微生物以及厚壁菌门(Firmicutes)的2个科的微生物。DGGE图谱亦显示了柑桔木虱内细菌类群受到柑桔木虱寄主植物和地理位置的影响而存在差异,其中寄主植物的影响比地理位置的影响更明显。合胞内共生菌Syncytium endosymbiont在柑桔木虱内稳定存在不受寄主和地理位置的影响。柑桔木虱内的合胞体内共生菌(Syncytium endosymbiont)是主要的优势微生物。PCR-DGGE和RFLP两种方法相结合,初步反应了柑桔木虱内细菌群落的多样性。

In this paper, The microbial diversity of citrus psyllids (Diaphorina citri) which vectors Liberobacter sp was been analyse, Using 16S rDNA-based molecular approaches of denaturing gradient gel electrophoresis(DGGE)and restriction fragment length polymorphism(RFLP). Bacterial taxa obtained by RFLP including Pseudomonadaceae, Enterobacteriaceae, Xanthomonadaceae, Burkholderiaceae, Rickettsiaceae, Rhizobiaceae. Syncytium endosymbiont and secondary symbiont of Diaphorina citri can be also obtained by RFLP. Bacterial taxa obtained by DGGE include four families of Proteobacteria and two families of Firmicutes. The result by PCR-DGGE analysis also revealed that the bacterial population of Diaphorina citri is affected by the host's food and the host's geographic location.And the impact of host's food is more obvious than that of geographic location. However, The existence of Syncytium endosymbiont in Diaphorina citri is not affected by the host's food and the host's geographic location. The Syncytium endosymbiont of Diaphorina citri is a dominant microflora and the microbial diversity in Diaphorina citri.
