通过体外实验研究黄芪多糖(astragalus polysaccharide,APS)对慢性粒细胞白血病(chronic myelocytic leukemia,CML)患者浆细胞样树突状细胞(plasmacytoid dendritic cells ,pDC)功能的影响,为进一步阐明黄芪多糖的免疫学活性及其在免疫治疗中的应用提供依据。从慢性粒细胞白血病患者外周血分离单个核细胞,免疫磁珠法去除pDC,加入0、50、100、200mg/L的黄芪多糖共孵育24小时,收集上清液,应用酶联免疫吸附试验检测pDC分泌的IFN-α、IL-6、TNF-α量的变化。本实验应用慢性粒细胞白血病患者外周血来源的pDC,通过体外实验证明了黄芪多糖能够提高治疗后的慢性期CML 患者pDC分泌的IFN-α、IL-6、TNF-α量及初诊未治慢性期CML 患者pDC分泌的IL-6、TNF-α量,并且呈黄芪多糖浓度依赖性。本实验结果证实了黄芪多糖能够增强慢性粒细胞白血病患者pDC的功能。

o explore the mechanism of immunomodulatory activity of Astragalus polysaccharide (APS) on chronic myelocytic leukemia peripheral blood mononuclear cells(PBMC)-derived plasmacytoid dendritic cells (pDC). Methods We isolated chronic myelocytic leukemia PBMC and isolated pDC by MACS, then incubated with APS (0、50、100、200mg/L )for 24 hours.We used ELISA to determine the effect of APS . Results We analyzed the concentration of IFN-α, IL-6,TNF-α secreted by pDC using ELISA.APS-treated pDC secreted higher level of IFN-α, IL-6,TNF-α than untreated pDC. Conclusion The result suggest that APS can promote the function of pDC from CML.
