二氧化硫对低氧性肺动脉高压大鼠右心室肥厚和肺血管炎症的调节作用

目的: 探讨二氧化硫(sulfur dioxide, SO2)对低氧性肺动脉高压发病过程中右心室肥厚的影响,及其对低氧性肺动脉高压大鼠肺血管炎症的调节作用及其机制。方法:26 只雄性Wistar 大鼠随机分为对照组8 只、低氧组8 只和低氧+SO2供体(Na2SO3/NaHSO3 溶液)组10 只。实验中采用常压低氧的方法,氧浓度为10%,每天低氧6 小时,共持续21 天。 低氧处理结束后,测量RV/(LV+SP)比值,Western blot方法检测三组大鼠肺组织中核转录因子-κB(nuclear factor-κB, NF-κB)和细胞间粘附分子(intracellular adhesion molecule-1, ICAM-1)的蛋白表达。结果:与对照组相比,低氧组大鼠右心室明显肥厚(P <0.01),肺组织中NF-κB和ICAM-1蛋白表达显著升高(P <0.01)。与低氧组相比,低氧+SO2 供体组大鼠右心室肥厚明显缓解(P<0.05),肺组织NF-κB蛋白和ICAM-1蛋白均显著降低(P<0.05或P <0.01)。结论:SO2可显著缓解低氧性肺动脉高压大鼠右心室肥厚,抑制低氧性肺动脉高压大鼠肺血管炎症反应。
Objective:The study was to investigate regulatory effects of sulfur dioxide on right ventricular hypertrophy and pulmonary inflammation in rats with hypoxic pulmonary hypertension as well as its mechanisms. Methods: 26 male Wistar rats were randomly divided into control group (n=8), hypoxic group (n=8) and hypoxic+SO2 group (n=10). Normobaric hypoxic treatment was used to rats with a O2 concentration of 10%, 6 h each day for 21 days. RV/(LV+SP)ratio,nuclear factor-κB (NF-κB) and intracellular adhesion molecule-1 (ICAM-1) were detected with Western blot. Results: Compared with that of control rats right ventricular hypertrophy developed(P <0.01) and lung tissue NF-κB and ICAM-1 expression markedly increased(P <0.01) in hypoxic rats. Compared with hypoxic rats,right ventricular hypertrophy obviously lessened and lung tissue NF-κB and ICAM-1 expression markedly reduced in rats of hypoxic+SO2 group (P<0.05). Conclusion: SO2 markedly lessened right ventricular hypertrophy and inhibited hypoxic pulmonary vascuilar inflammation in hypoxic pulmonary hypertensive rats.
