旋毛虫虫体蛋白诱导Hela细胞凋亡研究

目的:观察旋毛虫虫体蛋白对体外培养人宫颈癌Hela细胞的凋亡诱导作用,并阐明其作用机制。方法:通过体内、体外培养获得旋毛虫成虫与新生幼虫的混合物,采用匀浆、离心的方法制备旋毛虫虫体蛋白,分别用50μg/ml旋毛虫虫体蛋白和PBS处理体外培养的宫颈癌Hela细胞,观察细胞形态变化,Annexin V-EGFP/PI染色法检测细胞凋亡情况,RT-PCR、Western bloting方法检测survivin、caspase3和caspase9基因的转录与蛋白表达情况。结果:实验组细胞回缩呈圆形,边缘不清,部分细胞脱落悬浮;Annexin V-EGFP/PI时实验组细胞出现不同程度的绿染和红染,提示细胞出现凋亡;半定量RT-PCR法和Western blot法检测显示实验组caspase3、caspase9基因转录表达增加,survivin基因转录表达减少。结论:旋毛虫虫体蛋白可诱导人宫颈癌Hela细胞凋亡,其作用与survivin表达下调,caspase3、caspase9表达上凋有关。
Objective:To expound the apoptosis induction mechanism of Trichinella spiralis protein on Hela cells. Methods: Trichinella spiralis (TS) were cultured in vivo and in vitro. TS protein was prepared from adult worm and muscle larvae by homogenization and centrifugation, and Hela cells were treated with PBS and TS protein respectively. Apoptosis of Hela cells was tested by Annexin V-EGFP staining. The expression of survivin, caspase3 and caspase9 were analyzed by RT-PCR and Western Blot. Results: Cells in the experiment group was observed characteristics of apoptosis in morphology by microscope. Apoptosis was detected by the Annexin V-EGFP staining. The expression of survivin was down-regulated, caspase3 and caspase9 were up-regulated by semiquantitative RT-PCR and Western blot analysis. Conclusion: Trichinella spiralis protein successful induced apoptosis of Hela cells.
