以自育月季品种"花仙子"和7个现代月季品种为试材,对Ty1-copia类逆转座子的RNaseH-LTR序列进行克隆与分析。根据Ty1-copia类逆转座子RnaseH 3′端具有保守结构域这一特点,对RNaseH-LTR序列进行克隆,共分离出7条RNaseH-LTR序列。分析显示7条序列同源性范围在7.7%-56.3%;序列内存在TATA-box和CAAT-box启动元件及多种逆境胁迫相关调控元件;RNaseH-LTR序列在长度和碱基组成上都存在着高度的异质性,而且每条序列都存在终止密码子突变,说明终止密码子突变可能是造成Ty1-copia类逆转座子RNaseH-LTR异质性的原因之一。

Self-breeding rose variety R. "Flower faerie" and other seven modern rose varieties were as plant materials. RNase H-LTR sequences of Ty1-copia retrotransposons were cloned and analysed. According to the Ty1-copia retrotransposon RnaseH 3 'end had the characteristics of conserved domain. RNaseH-LTR sequences were cloned. Seven RNaseH-LTR sequences were isolated. The results showed that seven sequences homology were range from 7.7% to 56.3%. All the sequence existed TATA box, CAAT box start components and multiple stress related regulatory elements. There was high degree of heterogeneity in RNaseH-LTR sequence length and base composition and each sequence had codon mutation. It was indicated that termination codon mutation might be the reason of heterogeneity of Ty1-copia retrotransposons RNaseH-LTR.
