三种胶原膜理化性质以及其降解产物的成骨特性的研究

目的:探究不同来源的三种胶原膜的理化性质,如表面微观形态、孔隙率、机械性能、降解性、接触角及其降解产物的成骨作用。方法:扫描电子显微镜观察三种胶原膜的表面微观形态,利用PBS溶液浸泡测定其孔隙率,万能测试机测定胶原膜的拉伸强度及最大伸长率,胶原膜于PBS溶液、人工唾液、胶原酶溶液中降解,测定其降解速率,接触角测量仪测定胶原膜的接触角,比较其亲水性,通过细胞增殖和分化实验探究三种胶原膜降解后产物的成骨特性。结果:桀亚真皮和Bio-Gide的粗糙面可见胶原纤维排列疏松,吉特瑞表面并无胶原纤维;Bio-Gide的孔隙率和接触角相对更小,亲水性更强;桀亚真皮和吉特瑞的机械强度更大;桀亚真皮降解率最小;桀亚真皮和吉特瑞的降解产物对细胞增殖和分化作用更明显。结论:探究三种胶原膜不同的理化性质,各有其优势,临床中可根据相应的病例选择相应的胶原膜。

Objective: The aim of the present study was to evaluate the physicochemical properties of three kinds of collagen membranes from different sources, by evaluating surface topography, porosity, mechanical properties, degradability, contact angle and osteogenesis of degradation products. Methods: The surface topography of three kinds of collagen membranes was observed by scanning electron microscope. The porosity or swelling property was measured by PBS solution immersion.The tensile strength and elongation at break of collagen membranes were measured by universal testing machine.The collagen membranes were degraded in PBS solution, artificial saliva and collagenase solution to messure the degradation rate.The contact angle of collagen membrane was measured by contact angle measuring instrument, and its hydrophilicity was compared. The osteogenic characteristics of the degraded products of three kinds of collagen membrane were studied by cell proliferation and differentiation experiments. Results: the rough surface of Jieya dermis and Bio-Gide showed loose arrangement of collagen fibers, but there were no collagen fibers on the surface of Giteri; the porosity and contact angle of Bio-Gide were relatively smaller, and the hydrophilicity was stronger; the mechanical strength of Jieya dermis and Gitri was higher.The degradation rate of Jieya dermis was the lowest. The effect of cell proliferation and differentiation of degradation products of Jieya dermis and Giteri was more obvious. Conclusion: This paper studies the different physicochemical properties of three kinds of collagen membrane, each of which has its own advantages, and the corresponding collagen membrane can be selected according to the corresponding cases in clinic.
