油菜芥甘种间杂交来源的导入系RAPD分析
RAPD analysis of the rapeseed introgression lines derived from the interspecific cross Brassica juncea × Brassica napus
【研究目的】阐明油菜芥甘种间杂交的遗传规律、种间杂种后代的最佳选择世代以及芥菜型油菜遗传物质的渐渗与油菜育种需要的优异种质之间的关系。【方法】利用随机扩增多态性DNA(RAPD)标记对3个用于杂交的亲本和18个芥菜型油菜与甘蓝型油菜种间杂交来源的渐渗系(BC1F7代)进行了遗传分析。【结果】检测到芥菜型油菜亲本遗传物质在渐渗系中有不同程度保留,平均为13.5%,而两个甘蓝型油菜亲本的遗传物质明显高于芥菜型油菜,分别为50.8%和48.0%。通过对供试材料的遗传距离分析和聚类分析并结合系谱分析,发现芥甘种间杂交BC1F3代已基本稳定,此时应选留较多优异单株,以便获得更多优异种质。【结论】芥菜型油菜的遗传物质通过芥甘种间杂交已成功转育到甘蓝型油菜中,在芥甘种间杂交后代选育过程中BC1F3是一个重要的选择世代。
【OBJECTIVE】To clarify the genetic principles and the proper selection generation(s) of the interspecific cross between Brassica napus and B. juncea, and to reveal the relationship between introgression of B.juncea genetic material and germplasm required for B. napus breeding.【METHOD】The random amplified polymorphic DNA(RAPD) markers were used for genetic analysis of the three parents and the eighteen introgression lines (ILs, BC1F7) selected from the interspecific cross B. napus and B. juncea【RESULTS】B. juncea genetic material was maintained in the ILs at various degrees, with an average of 13.5% while the two B.napus parents contribute to 50.8% and 48.0% of genetic components in ILs, respectively. Combined analysis of genetic distance, clustering and pedigree showed that the BC1F3 was the generation when the interspecific progenies became inheritably stable and can be selected for superior individual plants.【CONCLUSION】The genetic material was successfully introgressed from B. juncea into B. napus by interspecific crossing, and the selection at the generation BC1F3 is crucial for the interspecific cross breeding of B. juncea × B. napus.
