目的:探讨GKN1对胃癌细胞生物学行为的影响,以及GKN1与TFF1之间的结构和功能关系。方法:利用脂质体转染的方法构建GKN1、TFF1单独和共转染的稳定胃癌AGS细胞系,用免疫荧光和免疫组化的方法观察二者在转染细胞内的定位;用免疫共沉淀的方法研究二者之间的结构关系。利用MTT、流式细胞技术、Tunel法和划痕实验研究GKN1和TFF1对胃癌AGS细胞系的增殖、凋亡、细胞周期以及细胞迁移的影响,并分析二者之间的功能关系。结果:免疫共沉淀结果显示,无论是在共转染胃癌细胞内还是正常胃黏膜上皮细胞中均未发现GKN1和TFF1存在直接结构上的相互作用。GKN1或TFF1均能够抑制转染胃癌细胞的增殖。但GKN1对胃癌细胞凋亡的促进更为明显。但二者对转染胃癌细胞迁移的影响不明显。GKN1和TFF1共转染时,胃癌细胞的增殖和凋亡并未出现二者作用的叠加或者协同效应。结论:GKN1具有抑制胃癌细胞增殖的作用,且对胃癌细胞凋亡的促进更为明显。GKN1和TFF1之间并不存在结构和功能上的相互作用。

im: To explore the effects of GKN1 on biological behaviours of gastric cancer cells, and to investigate the relationship between GKN1 and TFF1. Material and Methods:The structural interactions between GKN1 and TFF1 were further investigated by co-immunoprecipitation. The effects of GKN1 on gastric cancer AGS cell growth, cell cycle, apoptosis and migration were analyzed by MTT, Tunel, flow cytometry and scrape migration assay. The functional relationship between GKN1 and TFF1 were also explored. Results:The results of co-immunoprecipitation demonstrate that there was no direct structural interaction between GKN1 and TFF1. Both GKN1 and TFF1 could suppress the proliferation of transfected AGS cells, but had no effect on cell migration. Furthermore, GKN1 could significantly promote the apoptosis of transfected AGS cells. There was no functional relationship between GKN1 and TFF1 on the biological behavious of gastric cancer cells. Conclusions: GKN1 could suppress the proliferation and significantly promote the apoptosis of gastric cancer cells. However, no structural or functional relationship was found between GKN1 and TFF1.
