产淀粉酶海洋酵母菌多样性的研究
Research on Diversity of Amylase-Producing Marine Yeasts
从采集自南海、南极、烟台长岛、青岛近海的泥样和鱼类肠道内容物中分离得到了数百株海洋酵母菌,进一步筛选得到8株能够产淀粉酶的海洋酵母菌。根据对它们的生理和生化特征、所产粗酶最适pH和温度以及18S rDNA的部分序列的分析,确定菌株Ba、H、M和W1a属于解脂耶罗威亚酵母(Yarrowia lipolytica),菌株DMC属于平滑假丝酵母(Candida parapsilosis),菌株W13a、G7b和N13d属于普鲁兰短梗霉(Aureobasidium pullulans)。结果显示在海洋环境中产淀粉酶的海洋酵母菌多样性还是相当丰富的, 并发现解脂耶罗威亚酵母(Yarrowia lipolytica)、平滑假丝酵母(Candida parapsilosis)和普鲁兰短梗霉(Aureobasidium pullulans)是可以产生淀粉酶的新酵母菌菌株。
ifferent samples of sediments and guts of fish in the South China Sea, the South Pole, Changdao of Yantai and the coastal line of Qingdao were collected. Marine yeast strains from these samples were isolated and 8 strains amylase -producing marine yeasts were screened. Physiological and biochemical characters, optimum pH and temperature of the crude amylases produced by them were examined, and partial sequences of the 18S rDNA were also analyzed. Based on the physiological and biochemical characters and phylogenetic analyses, the eight strains were found to be closely related to Yarrowia lipolytica (Strain Ba, Strain H, Strain M and Strain W1a), Candida parapsilosis(Strain DMC), Aureobasidium pullulans (Strain W13a, Strain G7b and Strain N13d). These results indicated that amylase -producing marine yeasts were considerably diverse in the marine environments.
