合欢皮皂苷对小鼠毒性机制的研究
Study of toxicologic action and its mechanism of saponin from Albizia julibrissin Durazz on the mice
目的 研究合欢皮皂苷对小鼠毒性及机制。方法 采用一次性灌胃致毒剂量皂苷,观察14天时,合欢皮皂苷对小鼠肾毒性表现。HE染色观察病理形态的变化,生化法测定其对肾SOD和GSH-PX的影响,末端标记法检测肾细胞凋亡,免疫组织化学法检测肾组织Bcl-2、Bax、COX-1及COX-2基因蛋白的表达。 结果 合欢皮皂苷口服致毒剂量可引起肾组织形态的变化,降低肾SOD和GSH-PX的活性;肾小球内皮细胞凋亡数目增多,Bcl-2表达下降,COX-2表达增加;肾小管COX-1表达增加。结论 致毒量合欢皮皂苷对肾有一定毒性,促使细胞凋亡,可能与皂苷降低肾的抗氧化能力和改变凋亡相关基因的表达、升高COX-1的表达有关。
Objective To investigate the mechanism of total julibroside which caused the toxicity of kidney of mice. Methods After intragastric toxic doses of total julibroside for 14 days, the change of kidney was observed. The morphosis was observed by Hematoxylin Eosin staining. The activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX) was measured by biochemitrical method. The apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick-end labeling(TUNEL) method. The expression of COX-1 and COX-2 was detected by immu-histochemistry method. Results Toxic dose of total julibroside caused pathological changes in kidney, decreased the activity of SOD and GSH-PX, increased the number of apoptosis, reduced the expression of Bcl-2, increased the expression of COX-2 in epithelial cellsof Kidney Glomerulus and of COX-1 in kidney tubules. Conclusion It shows that total julibroside has toxicity to kidney. It perhaps related to increased apoptosis, decreased ability of anti-oxygen, reduced the expression of Bcl-2, and increased the expression of COX.
