焦核龙眼种子败育的cDNA-AFLP分析
cDNA-AFLP Analysis on Seed Abortion in Longan (Dimocarpus Longan Lour.)
采用 cDNA-AFLP 技术对龙眼正常和败育的种子进行了差异表达分析,并对差异明显的片段进行回收、克隆、测序以及利用 GenBank 数据库中已知序列进行 Blastn 和 Blastx同源性比对。结果表明:在 11 条重复性较好的差异片段中有 3 个 TDFs 分别与 Ca2+-ATP 酶、纤维素酶、质体 ATP/ADP 转运蛋白具有一定的相似性,这些基因片段的上调或下调表达可能影响了龙眼种子发育过程中的许多重要代谢途径,从而引起龙眼种子败育;4 个 TDFs 与一些 hypothetical 蛋白的序列相似;4 个 TDFs 在蛋白数据库中未找到匹配的序列,核酸数据库中比对的结果也显示较高 E-值,说明为非同源基因。
cDNA-AFLP technology was applied to identify diffierential genes expression between longan normal and abortive seeds. Transcript-derived fragments(TDFs) identied as dierentially expressed genes were eluted, cloned, sequenced and compared with database using BLAST at NCBI. The results showed that among 11 sequenced fragments, three TDFs presented similar with Ca2+-ATPase, cellulase, plastidic ATP/ADP transporter, respectively, whose up-regulated or down-regulated expression might influence several important metabolic pathways in the developing longan seeds and led to seed abortion; four TDFs were similar with hypothetical protein; other four TDFs were found no match to known protein and no significant similar with known nucleotide due to high E-value.
